Lipinski's Rule of Five



         


Christopher A. Lipinski formulated a so-called rule of 5 as criteria for the ability of a compound to be absorbed or permeate through biological barriers, the first of the ADME-Tox criteria for a potential drug. The rule derives its name from the fact that the relevant cutoffs are all multiples of 5:
